A Comparative Histochemical Study on the Mucosa of the Large Intestine


Abstract
A comparative histochemical study was done on the mucosubstance and alkaline phosphatase and acid phosphatase of the mucosa of the large intestine in the mouse, rat, guinea pig, rabbit, cat, dog and human fetus. The mucosubstance was observed with alcian blue staining(pH 2.5), PAS and combined staining of alcian blue(pH 2.5) and PAS. The alkaline and acid phosphatase were observed with Gomori¢¥s modified methods.
The results were as follows:
l. The surface and crypt columnar cells of the intestine were stained weakly with alcian blue and
with PAS, but negatively with combined staining of alcian blue and PAS in all the species.
2. The surface and crypt goblet cells of the intestine were stained moderately to strongly with alcian
blue and with PAS in all the species.
The surface and crypt goblet cells of the intestine were stained equally and strongly with alcian blue and PAS in combined staining in the mouse and dog, but moderately to strongly both with alcian blue and PAS, but more strongly with alcian blue than PAS in combined staining in the rat and rabbit.
4. The surface goblet cells of the intestine were stained equally and strongly with alcian blue and PAS, but the crypt goblet cells were stained strongly only with alcian blue in combined staining in the guinea pig.
5. The surface and crypt goblet cells of the lower part of the large intestine were stained equally and strongly with alcian blue and PAS in combined staining and the goblet cells of the upper part of the large intestine were stained strongly both with alcian blue and PAS, but more strongly with alcian blue than PAS in combined staining in the cat and human fetus.
6. The surface and crypt epithelial cells of the large intestine were strongly positive in alkaline phosphatase reaction in the cat, but the surface and crypt neck epithelial cells were moderately positive and the crypt base epithelial cells were weakly positive in alkaline phosphatase reaction in the dog. The surface epithelial cells were moderately positive but the crypt epithelial cells were almost negative in alkaline phosphatase reaction in the rabbit and guinea pig. The mucosa of the large intestine of the other animals and human fetus were almost negative in alkaline phosphatase reaction.
7. The surface and crypt epithelial cells of the large intestine were moderately to strongly positive in acid phosphatase reaction in the cat and weakly positive in acid phosphatase reaction in the caecum of the dog and in the colon of the guinea pig. The mucosa of the large intestine of the other animals and human fetus were almost negative in acid phosphatase reaction.
